บทคัดย่อ

ได้ศึกษาการชักนำ Xanthophyllomyces dendrorhous TISTR 5730 ให้กลายพันธุ์ด้วยการอาบนิวตรอนร่วมกับการฉายรังสี อัลตราไวโอเลตเพื่อเพิ่มประสิทธิภาพในการผลิตสารแคโรทีนอยด์ โดยนำ Xanthophyllomyces dendrorhous มาอาบ นิวตรอนที่นิวตรอนฟลักซ์ 10 10 นิวตรอนต่อตารางซม.ต่อวินาที เป็นเวลา 1 นาที จากนั้นนำมาเลี้ยงบน yeast malt extract agar ที่ 22 o ซ. เป็นเวลา 3 วัน แล้วนำมาทำการกลายพันธุ์ซ้ำด้วยรังสีอัลตราไวโอเลตความเข้ม 470 ไมโครวัตต์/ซม. 2 ระยะห่าง 10 นิ้ว เป็นเวลา 3 นาที ตรวจสอบลักษณะการเจริญและคัดเลือกเชื้อบน yeast malt extract agar ที่ผสมด้วย ? -ionone พบว่าสามารถเก็บเชื้อได้ 941 ไอโซเลท บนอาหารคัดเลือก และเมื่อนำไปหาค่าความคงตัวของเชื้อใน 5 รุ่น สามารถคัดเลือกเชื้อได้ทั้งหมด 5 ไอโซเลท คือ NU–195 NU–252 NU–525 NU–534 NU–742 และเมื่อนำเชื้อทั้ง 5 ไอโซเลทไปทดสอบคุณสมบัติในการผลิตสารแคโรทีนอยด์ โดยการเลี้ยงในอาหารเหลวyeast malt extract ที่อุณหภูมิ 22o ซ. เป็นเวลา 5 วัน แล้วนำมาสกัดด้วยอะซีโตนและปิโตรเลียมอีเทอร์ เพื่อหาปริมาณแคโรทีนอยด์ทั้งหมดโดยการวัดค่า การดูดกลืนแสงที่ 474 นาโนเมตร พบว่า NU– 252 เป็นสายพันธุ์กลายที่มีประสิทธิภาพในการผลิตสารแคโรทีนอยด์ได้สูงสุด เท่ากับ 106.49 ไมโครกรัม ต่อยีสต์ 1 กรัม ซึ่งสามารถผลิตได้สูงกว่าสายพันธุ์ตั้งต้น 1.65 เท่า

Abstract

Improvement of carotenoid production in Xanthophyllomyces dendrorhous TISTR 5730 by induced mutation with neutron rays combining with ultraviolet light was studied. Xanthophyllomyces dendrorhous was treated with neutron rays ( 10 10neutrons/cm 2/sec) for 1 minute and incubated on yeast malt extract agar at 22 oC for 3 days. The colonies were re-treated with ultraviolet radiation (470 ?w/cm 2) at the distance of 10 inches , for 3 minutes. 941 isolates were obtained following growth on selective medium supplemented with ? -ionone, and their stability was tested for 5 generations. There were only 5 highly-producing mutants, namely NU–195, NU–252, NU–525, NU–534 and NU–742. These mutants were tested for carotenoid production in yeast malt extract broth at 2 2 oC for 5 days. The cultures were extracted with acetone and petroleum ether. Total carotenoid contents in the petroleum ether extracts were determined by absorbance measurement at 474 nm. Analyses of five generations indicated that NU-252 produced the highest amount of total carotenoid, 106.49 ?g/g of yeast, which was 1.65 times that of the wild type.
